A 19-nor compound (19-nortestosterone derivative) is any AAS that lacks the methyl group at carbon position 19 (C-19) of the steroid nucleus. This seemingly minor structural change — removing a single methyl group — has profound pharmacological consequences that define the entire class.
In testosterone, the C-19 methyl group is required for aromatase (CYP19A1) to initiate the oxidative ring A aromatization reaction. Remove C-19 and aromatization becomes impossible — except in nandrolone, where some aromatization to estrone still occurs through alternative pathway. Trenbolone's additional Δ9,11 double bonds completely block even this residual pathway.
| Compound | Additional Modifications | A:A Ratio | Aromatizes? | 5α-Reduction | PR Activity |
|---|---|---|---|---|---|
| Nandrolone (DECA/NPP) | 19-nor only | 125:37 | Minimal → E1 | → DHN (inactive) | ~20% P4 |
| Trenbolone (Ace/E) | 19-nor + Δ9,11 | 500:500 | None | None (Δ11 blocks) | ~60% P4 |
| MENT (Trestolone) | 19-nor + 7α-methyl | 650:250 | → 7α-Me-E2 | → 7α-Me-DHN (active) | Strong |
The mechanism: 5α-reductase converts testosterone to DHT (higher AR affinity → amplifies androgenicity in androgen-sensitive tissues). 5α-reductase converts nandrolone to 5α-dihydronandrolone (DHN) — which has LOWER AR binding affinity than nandrolone. So 5α-reductase activity actually reduces nandrolone's androgenicity in tissues with high 5α-reductase expression (prostate, skin, scalp). This produces the dramatically low androgenic ratio of 37.
Trenbolone's Δ9 and Δ11 double bonds completely prevent 5α-reduction (the Δ11 bond prevents the enzyme from accessing the ring). Without 5α-reduction-mediated androgenicity attenuation in sensitive tissues, Trenbolone's full AR binding potency (~3–5× testosterone) is expressed uniformly — producing 500 androgenic and 500 anabolic ratings.
